Product Specific Information
The E11/19 monoclonal antibody binds to phosphoserine 349 corresponding to the 22 C-terminal amino acid residues of human CCR5. CCR5 is a receptor for several inflammatory CC-chemokines including CCL3 (MIP-1-alpha), CCL4 (MIP-1-beta), and RANTES.
Target Information
Human immunodeficiency virus (HIV) and related viruses require coreceptors, in addition to CD4, to infect target cells. Several G protein-coupled receptors such as CCR5, CXCR4, CCR3, CCR2b, and CCR8 have been identified as HIV coreceptors. Among them, CCR5 (CC-CKR-5) is a principal coreceptor for macrophage- and dual-tropic HIV-1 strains, mediating fusion and entry of human white blood cells.
CCR5 is essential for infection by HIV-1, HIV-2, and SIV. Beta-chemokines such as RANTES, MIP-alpha, and MIP-beta are ligands for CCR5 and inhibit infection by M-tropic HIV-1. CCR5 associates with the surface CD4-gp120 complex of HIV, leading to membrane fusion and viral entry into target cells. The amino-terminal domain and extracellular loops of CCR5 serve as HIV binding sites. CCR5 mRNA is expressed in lymphoid organs and monocytes.
