Product Specific Information

The MP1-22E9 antibody reacts with mouse granulocyte/macrophage colony-stimulating factor (GM-CSF) and functions as a neutralizing antibody. Mouse GM-CSF is a 14 kDa factor produced primarily by activated T cells and macrophages, but also by endothelial cells and fibroblasts in response to TNF-alpha, IL-2, IL-1, and IFN-gamma. GM-CSF promotes the growth of macrophages, granulocytes, and dendritic cells, and exists both as a membrane-bound form and as a complex with the extracellular matrix. Non-glycosylated GM-CSF is biologically active.

Applications Reported:
This antibody has been used for ELISA capture, intracellular staining, flow cytometry, neutralization assays, western blotting, and immunohistochemistry on frozen sections.

Applications Tested:
Validated as the capture antibody in sandwich ELISA for mouse GM-CSF detection, using biotin-labeled MP1-31G6 (13-7332) for detection and recombinant mouse GM-CSF (14-8331) as the standard. Recommended capture concentration: 1–4 µg/mL. Include a standard curve (1000 pg/mL–8 pg/mL) in each ELISA plate.

Target Information

The GM-CSF gene encodes a cytokine regulating macrophage and granulocyte production, differentiation, and function. The active protein exists as a homodimer in the extracellular space. Located at chromosome region 5q31, this gene cluster (including IL-4, IL-5, IL-13) is associated with 5q- syndrome and acute myelogenous leukemia. GM-CSF contributes to tissue inflammation and elevated levels are observed in SARS-CoV-2 patients with acute respiratory distress syndrome. Mice lacking GM-CSF or its receptor develop pulmonary alveolar proteinosis.
