Target Information
The protein tyrosine phosphatase PTPN22 (also known as PTP22, LYP, PEP, formerly PTPN8) is a genetic variant associated with susceptibility to autoimmune diseases such as type 1 diabetes and rheumatoid arthritis.
A missense SNP in PTPN22 encodes a hematopoietic-specific protein tyrosine phosphatase known as Lyp. The risk allele is found in approximately 17% of the general Caucasian population and 28% of Caucasian individuals with rheumatoid arthritis.
This variant may disrupt the P1 proline-rich motif important for interaction with the SH3 domain of CSK, potentially altering normal regulation of T cell activation. PTPN22 is thought to function as an effector and/or regulator of CSK in T cells and other hematopoietic cells.
