Target Information
Semaphorins are the largest family of axon guidance cues, divided into eight classes (classes 3–7 found in vertebrates).
- Class 3 Semaphorins: Secreted proteins
- Classes 4–6: Transmembrane proteins
- Class 7: Membrane-associated via glycosylphosphatidylinositol (GPI) linkage
They share a conserved ~400 amino acid sema domain and function as collapsing factors and mediators of axon repulsion, though they can also act as chemoattractants depending on context. Semaphorins play roles in cardiovascular development and immune cell antigen presentation regulation. Receptors mediating semaphorin signaling include Neuropilin and Plexin family proteins.
