Target Information
Glycogen synthase kinase-3 (GSK3)는 원래 glycogen synthase의 조절자로 확인된 단백질 키나아제로, glycogen 대사뿐 아니라 단백질 합성, 세포 증식, 분화, 미세소관 조립/해체, 세포사 등 다양한 세포 기능 조절에 관여합니다.
GSK3의 기질 특이성은 독특하며, 기질의 인산화는 GSK 인산화 부위로부터 네 개의 잔기 C-말단에 phosphoserine 또는 phosphotyrosine이 존재할 때만 일어납니다.
GSK3에는 α와 β 두 가지 isoform이 존재하며, 인산화를 통해 엄격히 조절됩니다.
GSK3β의 Ser9(또는 GSK3α의 Ser21) 인산화는 PKB에 의해 매개되며, 이로 인해 효소가 불활성화됩니다. 반면 Tyr216(또는 GSK3α의 Tyr279) 인산화는 GSK3의 활성화를 유도합니다.
활성화된 GSK3β는 p53, c-Myc, c-Jun, HSF-1, cyclin D1 등 다양한 단백질을 인산화하며, tau 단백질의 비정상적 인산화에도 관여하여 알츠하이머병의 진행에 중요한 역할을 합니다.
