Description:
The PC10 antibody recognizes proliferating cell nuclear antigen (PCNA), a 36 kDa protein also known as polymerase delta auxiliary protein. It reacts with human, mouse, and rat PCNA, showing peak expression during the S-phase of the cell cycle.

Applications Reported:
Reported for use in flow cytometry, immunoprecipitation, immunoblotting (WB), and immunohistochemistry.

Applications Tested:

• Flow cytometry: ≤0.25 µg/test (in 100 µL final volume; 10⁵–10⁸ cells/test)
• Immunocytochemistry (methanol-fixed MDCK cells): ≤5 µg/mL
  It is recommended to titrate the antibody for optimal performance.

Target Information

PCNA (polymerase delta auxiliary protein) is essential for DNA replication and involved in DNA excision and mismatch repair. It binds to CDK inhibitor p21, endonucleases Fen1 and XPG, and DNA cytosine 5-methyltransferase (MCMT).
PCNA serves as a marker for proliferating cells and tumor proliferation status. It is localized in the nucleus, forms a homotrimer, and increases DNA polymerase δ processivity during replication. Upon DNA damage, PCNA is ubiquitinated and participates in the RAD6-dependent repair pathway.
Two transcript variants encoding the same protein and pseudogenes on chromosome 4 and X have been identified.
