Description:
The 14H1 monoclonal antibody recognizes human Musashi-1 (MSI-1), a 39 kDa RNA-binding protein expressed in CNS stem and progenitor cells, with decreased expression in differentiating cells. Musashi-1 functions as a translational repressor regulating target proteins such as Numb and p21 (CIP1). It is expressed in various stem cells including gut, stomach, mammary gland, and hair follicles. Expression has also been identified in tumor types such as pulmonary and adenocarcinoma, and large/small cell carcinomas. Expression in ischemic lesions after stroke suggests roles in neurogenesis and apoptosis regulation.

Musashi-1 is highly conserved, and the 14H1 antibody recognizes mouse and rat Musashi-1 as well.

Applications Reported:
This antibody has been reported for use in immunoblotting (WB), immunohistology of paraffin-embedded sections, and immunocytochemistry.

Applications Tested:
Tested by immunofluorescent staining of paraformaldehyde-fixed, permeabilized cells. Recommended for use ≤10 µg/mL. Optimization by titration is advised.

Target Information

Musashi-1 belongs to the Musashi family of RNA-binding proteins involved in cell fate determination and maintenance of stem-cell states. These proteins regulate gene expression post-transcriptionally and contain two conserved RNA recognition motifs. In mammals, Musashi-1 is expressed in fetal kidney, brain, liver, and lung, and in adult brain and pancreas. In humans, the gene is located on chromosome 12.
