Product Specific Information

This antibody reacts with SLP76, a 76 kDa cytosolic adaptor protein involved in signaling of various hematopoietic cells such as T cells, mast cells, and neutrophils. In B cells, it is replaced by SLP65.

Target Information

SLP-76 was originally identified as a substrate of the ZAP-70 protein tyrosine kinase following T cell receptor (TCR) ligation in the leukemic T cell line Jurkat. The SLP-76 locus is located on human chromosome 5q33, and the gene structure has been partially characterized in mice.
The human and murine cDNAs encode 533 amino acid proteins that are 72% identical and consist of three modular domains:

• NH₂-terminus: Contains an acidic region with a PEST domain and several tyrosine residues phosphorylated following TCR ligation
• Central domain: Proline-rich region
• COOH-terminus: SH2 domain

SLP-76 interacts with several proteins constitutively and inducibly following receptor ligation, functioning as an adaptor or scaffold protein. Studies using SLP-76 deficient T cell lines or mice demonstrate that SLP-76 plays a positive role in T cell development, activation, and mast cell and platelet function.
