Product Specific Information

This antibody recognizes an extracellular epitope on native surface-expressed HLA-E, but not the denatured heavy chain of HLA-E.
HLA-E is a nonclassical MHC Class I molecule (MHC Class Ib) expressed on many types of human cells.
The antibody shows remarkable specificity for HLA-E, with only weak cross-reactivity to classical MHC Class I molecules (MHC Class Ia): HLA-A24, -B7, -B27, -B51, -B54, -C7.

Target Information

HLA-E belongs to the HLA class I heavy chain paralogues.
This class I molecule is a heterodimer consisting of a heavy chain and a light chain (beta-2 microglobulin).
The heavy chain (~45 kDa) is membrane-anchored and encoded by 8 exons:

1. Exon 1: leader peptide
2. Exons 2–3: α1 and α2 domains (peptide binding)
3. Exon 4: α3 domain
4. Exon 5: transmembrane region
5. Exons 6–7: cytoplasmic tail

HLA-E binds a restricted subset of peptides derived from the leader peptides of other class I molecules.
HLA and MHC antibodies are key tools in immunopeptidomics, aiding in the identification and characterization of neoantigens via high-performance liquid chromatography coupled to tandem mass spectrometry (LC–MS/MS).
