This antibody reacts with an extracellular epitope of CD16, a low affinity receptor for aggregated IgG (FcgammaRIII antigen).
CD16 exists in two isoforms:
- CD16a (FcgammaRIIIA): 50–65 kDa, expressed on NK-cells, monocytes, macrophages
- CD16b (FcgammaRIIIB): 48 kDa, mainly expressed on neutrophils
Target Information
CD16 encodes a receptor that recognizes the Fc portion of immunoglobulin G and is involved in immune complex clearance and other functions such as cellular mediated cytotoxicity and enhancement of virus infections.
The gene FCGR3A shares high similarity with FCGR3B, located on chromosome 1.
FCGR3A is expressed on NK cells as an integral membrane glycoprotein, while FCGR3B is expressed on polymorphonuclear neutrophils (PMN) anchored through a phosphatidylinositol (PI) linkage.
Mutations in this gene have been associated with Immunodeficiency 20, susceptibility to recurrent viral infections, systemic lupus erythematosus, and alloimmune neonatal neutropenia.
Alternatively spliced transcript variants encoding different isoforms have been identified.
