Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It plays a key role in T cell signal transduction, differentiation, and activation. CD8 is mainly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with the T cell receptor (TCR) for MHC class I molecules. In contrast, CD4 acts as a co-receptor for MHC class II molecules.

CD8 is also expressed on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8α homodimers. Upon binding to MHC-I/peptide complexes on antigen-presenting cells, CD8 recruits Lck kinase, promoting lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs play an essential role in pathogen and tumor cell clearance. Differentiation of naïve CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
