Target Information
IL-21 is a 17 kDa immunomodulatory cytokine produced mainly by Natural Killer (NKT), T helper (Th) 17, and T follicular helper (TFH) cells. In TFH cells, IL-21 expression leads to autocrine signaling through IL-21R and STAT3, resulting in transcriptional activation by Bcl6.
Similar to IFN-γ for Th1 and IL-4 for Th2, IL-21 is essential for TFH cell development and effector function. It promotes T cell-dependent B cell differentiation into plasma and memory cells, stimulates IgG production, and induces apoptosis in naive B cells.
In Th17 cells, IL-21 expression with STAT3, IRF4, and RORγt upregulates IL-23R, promoting Th17 maturation and maintenance by IL-23. IL-21 also downregulates Foxp3 expression.
IL-21 deficient mice show resistance to chemically induced colitis due to impaired Th17 molecule upregulation, while elevated IL-21 levels are observed in colitis models.
