Description:
The monoclonal antibody OX110 recognizes mouse CD200R (OX2). CD200R is an inhibitory receptor structurally similar to its ligand CD200 but lacks a typical ITIM domain. Instead, it contains a phosphotyrosine binding domain (NPXY) that, upon binding SHIP, inhibits ERK, JNK, and MAPK p38 pathways. In activated macrophages, this signaling pathway inhibits TNFα secretion.

Expression:
CD200R expression is restricted to hematopoietic cells, including myeloid cells (monocytes, macrophages, DCs, neutrophils, mast cells, basophils), a subset of lymphocytes, Langerhans cells, and dendritic epidermal T cells.

Applications Reported:
This OX110 antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of Fc-blocked mouse thioglycolate-elicited peritoneal cells. Recommended use ≤ 0.25 µg/test (100 µL final volume). Optimal titration should be determined empirically (10⁵–10⁸ cells/test).

Target Information

CD200R (OX2R) is a receptor for the OX-2 membrane glycoprotein. Both receptor and ligand are cell surface glycoproteins with two immunoglobulin-like domains. CD200R is mainly expressed on myeloid lineage cells and functions as a myeloid downregulatory signal. In mice, CD200R is found primarily on monocytes and neutrophils, as well as T-lymphocytes and mast cells. Alternative splicing results in multiple transcript variants. Dysfunction of CD200R is associated with primary hypertrophic osteoarthropathy.
