Description:
The P3TUNYA monoclonal antibody reacts with mouse CD218a (Interleukin 18 Receptor alpha, IL-18Ra). CD218a forms part of the heterodimeric receptor complex for IL-18, a proinflammatory cytokine in the IL-1 family. IL-18 is synthesized as an inactive precursor requiring caspase cleavage for activation and is secreted by activated monocytes, macrophages, epithelial cells, and keratinocytes.

IL-18 Receptor is expressed on Th1 cells, subsets of NK cells, and neutrophils, but not on naive CD4 T cells or Th2 cells. The receptor consists of a ligand-binding alpha chain (IL-18Ra) and a signal-transducing beta chain (IL-18Rb). IL-18Ra alone binds IL-18 with low affinity, which increases upon complex formation with IL-18Rb. IL-18R signaling is crucial for Th1 differentiation and effector function, inducing IFN-γ, TNF-α, and Fas ligand expression in T and NK cells. IL-18 also synergizes with IL-23 in Th17 cell development and function.

A subset of CD8 T cells (Tc17) co-secretes IFN-γ with IL-17A and sometimes IL-22, sharing features with Th17 cells. In humans, this population expresses CD161 and IL-18R. Since a CD161 homolog is not identified in mice, IL-18R may serve as a surrogate marker for CD161 in mouse Tc17 phenotyping.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes at ≤0.25 µg/test. A test is defined as the amount of antibody sufficient for staining a cell sample in 100 µL final volume. Optimal titration is recommended for specific assay conditions.

Target Information

IL18R1 (Interleukin 18 Receptor 1) is a cytokine receptor belonging to the interleukin 1 receptor family. It specifically binds IL-18 and is essential for IL-18–mediated signal transduction. Expression is induced by IFN-α and IL-12 in NK and T cells. The IL18R1 gene forms a cluster on chromosome 2q with IL1R2, IL1R1, ILRL2 (IL-1Rrp2), and IL1RL1 (T1/ST2).
