Product Specific Information

Monoclonal antibody 42D1 recognizes the CD85d (ILT4/LIR-2) protein, a member of the ILT/LIR/MIR receptor family. CD85d is a 110 kDa transmembrane receptor with three Ig-like domains and a long cytoplasmic tail containing ITIM motifs. It is expressed on monocytes, macrophages, and dendritic cells, and binds non-classical class I HLA molecules (HLA-G, -B, G1). Upon ligand binding, SHP-1 interacts with ITIM, contributing to immune tolerance during fetal implantation and potentially influencing organ allograft rejection.

Crosslinking with the 42D1 antibody inhibits calcium flux, enhances HLA-G tetramer staining, and restores stimulatory activity of tolerogenic dendritic cells.

Target Information

LILRB2 (ILT4) encodes a leukocyte immunoglobulin-like receptor located at chromosome 19q13.4. The protein contains multiple extracellular Ig domains, a transmembrane region, and cytoplasmic ITIM motifs. Expressed on immune cells, it binds MHC class I molecules, transmitting inhibitory signals that modulate immune activation and prevent autoreactivity. Multiple transcript variants exist, encoding different isoforms.
