Description:
The AREG559 monoclonal antibody reacts with human Amphiregulin (AR), a member of the Epidermal Growth Factor (EGF) family. AR is expressed as a transmembrane precursor protein and released by proteolytic cleavage. Soluble AR binds to the EGF receptor (EGFR), promoting growth of epithelial cells, fibroblasts, and keratinocytes.
In the immune system, AR functions in tissue repair, homeostasis, infection tolerance, and inflammation suppression. It is expressed in T cells, group 2 innate lymphoid cells, basophils, mast cells, eosinophils, and neutrophils under various conditions.

Applications Reported:
This AREG559 antibody is validated for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometry on stimulated normal human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and the BestProtocols® “Protocol A: Two-step protocol for intracellular proteins.”
Recommended use: 5 µL (0.5 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.
Excitation: 633–647 nm; Emission: 660 nm; Laser: Red Laser.

Target Information

Amphiregulin (AR) binds to EGFR with lower affinity than EGF. The mature secreted AR is an 84-amino acid glycosylated growth regulator derived from a 252-amino acid transmembrane precursor.
EGFR ligands include EGF, TGFα, AR, HB-EGF, cripto-1, epiregulin, and betacellulin, all containing a conserved EGF-like domain with six cysteine residues forming three disulfide-bonded loops.
AR protein is localized in the epithelium of the colon, stomach, pancreas, breast, and placenta, and is overexpressed in cancers of the breast, colon, stomach, and pancreas.
