Target Information

Progression through the cell cycle requires activation of cyclin-dependent kinases (Cdks). Cdk2, a critical enzyme for initiation of cell cycle progression, is inactive alone but partially activated by cyclins such as cyclin D1 and fully activated by phosphorylation at Thr160. The enzyme responsible for this phosphorylation, Cdk-activating kinase (CAK), consists of a catalytic subunit, a regulatory subunit, and a subunit of unknown function. The regulatory subunit, cyclin H, is required for activation of Cdk7. Mat1, now identified as a protein associated with the cyclin H–Cdk7 nuclear complex throughout the cell cycle, facilitates the formation of an active Cdk-cyclin complex and exhibits kinase activity towards Cdk activation domains and RNA polymerase II.
