Target Information
Myelin protein zero (MPZ) is a Type I integral membrane glycoprotein that mediates adhesion of the myelin sheath layers, ensuring stable synaptic transmission. MPZ represents about 50% of total protein in the peripheral myelin sheath, contributing to structural integrity.
MPZ facilitates compact myelin wrapping through glycine zipper interface-dependent dimer and tetramer formation. Mutations such as G134R can disrupt multimer formation, leading to demyelinating neuropathies including Charcot-Marie-Tooth disease.
The cytoplasmic domain undergoes serine and tyrosine phosphorylation, especially during peak nerve myelination. MPZ transcript levels are moderate in brain, abundant in thymus, and highest in white matter of the CNS.
