Product Specific Information
- IHC (PFA fixed): Incubate antibody for 30 minutes at room temperature.
- Formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
IL-1 alpha (Interleukin-1 alpha) is a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. It is encoded by the IL1A gene and signals through receptors IL-1RI and IL-1RII, shared with IL-1 beta. Located on chromosome 2q13, IL-1 alpha plays a key role in innate immune defense by inducing other proinflammatory cytokines and initiating acute-phase responses.
IL-1 alpha activity is regulated by IL-1 Receptor Antagonist (IL-1RA), which inhibits receptor binding competitively. It influences NF-kappaB and MAP kinase pathways, stimulates IL6 and COX-2 expression, and enhances nitric oxide production. Elevated IL-1 alpha levels are associated with chronic inflammatory diseases such as rheumatoid arthritis, psoriasis, and multiple sclerosis.
