Product Specific Information
The EM92 monoclonal antibody reacts with mouse and human Oct3/4, encoded by the Pou5F1 gene. Oct3/4 is a POU domain-containing transcription factor critical for maintaining embryonic stem (ES) and induced pluripotent stem (iPS) cells in a pluripotent state. It is expressed by ES cells, embryonic germ cells, and embryonic carcinoma cell lines.
In inner cell mass (ICM) cells, reduction of Oct3/4 expression causes dedifferentiation to trophoectoderm, while increased expression leads to differentiation into mesoderm and primitive endoderm. Oct3/4 regulates genes such as FGF-4, UTF1, Sox2, Fbx15, Rex1, and osteopontin, often synergizing with Sox2 in gene regulation.
Oct3/4 expression can be downregulated in ES cells by retinoic acid treatment or by removal of leukemia-inhibitory factor (LIF).

Applications Tested: Western blot analysis of F9 and P19 embryonal carcinoma cell lysates. Recommended starting dilution: 2 µg/mL (optimize per assay).
Target Information
POU5F1 (Oct-4) is a maternally expressed octamer-binding transcription factor essential for early development. It plays a key role in maintaining pluripotency and has been instrumental in the generation of induced pluripotent stem (iPS) cells.
iPS cells can be generated by expressing POU5F1, Sox2, Klf4, Lin28, and c-Myc in fibroblasts. Recent studies show iPS cells can also be created using plasmid-based expression systems, eliminating viral vectors and improving safety for regenerative medicine applications.
