Product Specific Information

The monoclonal antibody 24B72D11 recognizes mouse and rat Mammalian achaete-scute homolog 1 (MASH1), a 34 kDa basic helix-loop-helix (bHLH) transcription factor. MASH1, similar to Olig2 and Ngn2, is expressed in progenitor cells of the nervous system and retina and regulates cell fate specification.
Expression of MASH1 is controlled by the Delta/Notch1 pathway and involves Hes1 binding as well as notch-mediated regulation of sox2, which upregulates MASH1 transcription.
MASH1 participates in the formation of dopaminergic, serotonergic, glutaminergic, and GABAergic neurons and is overexpressed in small cell lung carcinoma and medullary thyroid cancer.

Applications Reported: Western blotting, immunohistochemical staining (frozen tissue), and immunocytochemistry.
Applications Tested:

  • Western blot on reduced PC12 cells ≤ 5 µg/mL
  • Immunocytochemistry on fixed and permeabilized PC12 cells ≤ 20 µg/mL
    Antibody titration is recommended for optimal performance.

Target Information

ASCL1 (also known as ASH1) is a bHLH transcription factor essential for early nervous system development. It is expressed in fetal brain and required for the development and survival of autonomic neurons.
ASCL1 interacts with MEF-2A to regulate genes involved in neuronal circuit formation.
High ASCL1 expression is observed in neuroendocrine tumors, including medullary thyroid cancer and small cell lung cancer, serving as a potential marker for neuroendocrine differentiation.
The ASCL1 gene maps to human chromosome 12 and includes a trinucleotide repeat region, making it a candidate for inherited disorders.
