Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
CD146 (MCAM) is an integral membrane glycoprotein of the immunoglobulin superfamily. It is expressed at cell-cell junctions in endothelial cells and other normal or malignant cell types.
CD146 functions as a calcium-independent cell adhesion molecule and may act in signal transduction by recruiting Fyn kinase, leading to tyrosine phosphorylation of intracellular proteins involved in actin cytoskeleton assembly.
It plays a role in endothelial cohesion and melanoma metastasis through vascular interaction. Additionally, CD146 is active in neural crest cells during embryonic development and triggers tyrosine phosphorylation of FYN and PTK2, with a transient increase in intracellular calcium concentration.
