• Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C followed by cooling at room temperature for 20 min.

Target Information

CD146 (MCAM) is an integral membrane glycoprotein belonging to the immunoglobulin superfamily. It is expressed at cell-cell junctions in endothelial cells and other normal and malignant cell types.
CD146 functions as a calcium-independent cell adhesion molecule and may also participate in signal transduction by recruiting Fyn kinase, leading to tyrosine-phosphorylation of intracellular proteins involved in actin cytoskeleton assembly.
It plays a key role in cell adhesion and endothelial cohesion, and may facilitate melanoma cell interaction with vascular elements, enhancing tumor spread. Additionally, CD146 may act as an adhesion molecule in neural crest cells during embryonic development and trigger intracellular calcium increase through FYN and PTK2 phosphorylation pathways.
