• Immunohistochemistry (PFA fixed):
  Incubate antibody for 30 min at room temperature.
  Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD146 (MCAM) is an integral membrane glycoprotein belonging to the immunoglobulin superfamily. It is expressed at cell-cell junctions in all endothelial cells, and also in various normal and malignant cell types.
In addition to its role as a calcium-independent cell adhesion molecule, CD146 may function in signal transduction through recruitment of Fyn kinase, leading to tyrosine phosphorylation of intracellular proteins involved in actin cytoskeleton assembly.
CD146 contributes to endothelial cohesion and may facilitate melanoma cell interaction with vascular elements, enhancing hematogenous tumor spread. It may also function as an adhesion molecule in neural crest cells during embryonic development and act as a surface receptor triggering phosphorylation of FYN and PTK2 with transient increases in intracellular calcium.
