Target Information
Protein p21 is a tumor suppressor protein that inhibits Cyclin-dependent kinases (CDK) and is transcriptionally activated by p53. It plays a key role in the cellular response to genotoxic stress and is a major transcriptional target of p53. Nuclear p21 binds and inhibits Cdk1 and Cdk2, blocking cell cycle progression from G1 to S phase or G2 to mitosis after DNA damage, enabling DNA repair. In the cytoplasm, p21 exerts anti-apoptotic effects by inhibiting caspase 3 and apoptotic kinases ASK1 and JNK. p21 has dual roles in carcinogenesis, functioning both as a tumor suppressor and oncogene.
