Target Information
Protein p21 is a tumor suppressor protein that inhibits Cyclin-dependent kinases (CDK) and is transcriptionally activated by p53. It plays a critical role in cellular response to genotoxic stress and acts as a major transcriptional target of p53.
In the nucleus, p21 binds and inhibits Cdk1 and Cdk2, blocking the G1–S and G2–M transitions after DNA damage to allow DNA repair. In the cytoplasm, p21 has anti-apoptotic effects by inhibiting caspase 3 and apoptotic kinases ASK1 and JNK.
Thus, p21 exhibits dual functions in carcinogenesis, acting both as a tumor suppressor and oncogene depending on context.
