Product Specific Information

The J105 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the CD28 immunoglobulin superfamily. PD-1 contains an immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune regulation. It is expressed predominantly on activated T and B lymphocytes.
Two ligands, PD-L1 (B7-H1) and PD-L2 (B7-DC), interact with PD-1 and regulate immune responses through inhibitory signaling.

Costaining experiments indicate that eBioJ105 recognizes a different epitope than MIH4 (cat. 11-9969-42).

Applications Reported:

• Flow cytometric analysis.

Applications Tested:

• Pre-diluted and validated for flow cytometry using stimulated normal human peripheral blood cells.
• Recommended usage: 5 µL (0.5 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Ultra Violet 805™ emits at 797 nm and is designed for cytometers equipped with a 355 nm UV laser. Ensure instrument compatibility before use.

Staining Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies (Super Bright, Brilliant Violet, Brilliant Ultra Violet), use Super Bright Complete Staining Buffer (Product # SB-4401) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific polymer interactions.

Light Sensitivity:
This tandem dye is sensitive to light-induced oxidation. Protect from light during storage and use.

Fixation:
Samples may be fixed with IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.
Brilliant Ultra Violet 805™ is compatible with short-term methanol fixation (<1 hour).

Target Information

PD-1 (PDCD-1) is an inhibitory receptor expressed on activated T cells. It binds to PD-L1 and PD-L2 on antigen-presenting cells, transmitting inhibitory signals that regulate immune activation.
PD-1 knockout mice exhibit hyperactivated immune systems and autoimmune phenotypes.
Although PD-1 has a predicted molecular weight of 55 kDa, it often migrates at a higher molecular weight in SDS-PAGE.
