• Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
• For formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, then cool at room temperature for 20 minutes before staining.

Target Information

CD103 (ITGAE, Integrin alpha E) is expressed on mucosa-associated and activated T lymphocytes, as well as a subset of TGF-β1 cells.
It consists of a 150 kDa alpha chain and a 120 kDa beta chain. CD103 is expressed by approximately 0.5–2% of resting lymphocytes in peripheral blood and lymphoid organs.
The antibody stains a few bone marrow cells and, rarely, peripheral blood lymphocytes.
CD103 belongs to the integrin family, which are heterodimeric integral membrane proteins composed of alpha and beta chains.
The ITGAE gene encodes an I-domain-containing alpha integrin that undergoes post-translational cleavage, forming disulfide-linked heavy and light chains.
In combination with the beta 7 integrin, CD103 forms the E-cadherin binding integrin known as the human mucosal lymphocyte-1 antigen.
CD103 is preferentially expressed in human intestinal intraepithelial lymphocytes (IEL) and may serve as an accessory molecule for IEL activation.
Diseases associated with CD103 dysfunction include hairy cell leukemia and splenic marginal zone lymphoma.
