Product Specific Information
The eBioTB2F antibody reacts with Transforming Growth Factor-beta (TGF-beta), a pleiotropic cytokine existing in five isoforms (TGF-beta1–5). Isoforms share 70–80% homology but none with TGF-alpha.
TGF-beta1 is the most abundant form, widely expressed in lymphoid organs. All isoforms are biologically active as disulfide-linked homodimers, with monomeric subunits of 112 amino acids. Isoforms are derived from proteolytic cleavage of precursor proteins and are highly conserved across species (≈98% identity).
TGF-beta1 is produced at high levels by platelets and also by macrophages, lymphocytes, endothelial cells, chondrocytes, and leukemic cells. Its secretion can be induced by steroids, retinoids, EGF, NGF, vitamin D3, and IL-1.
It inhibits cell growth of epithelial cells, fibroblasts, neurons, lymphoid cells, and hematopoietic cells, suppresses T cell and NK cell proliferation, and downregulates macrophage activity.
TGF-beta1 also blocks IL-2–mediated anti-tumor activity of LAK cells.
Recent findings show TGF-beta1 plays a critical role in regulatory T cell development. Tumor-exposed dendritic cells secrete TGF-beta1, promoting natural Treg expansion and Foxp3 expression, leading to differentiation of CD4+CD25+ Treg cells (Ti-Treg).
Applications Reported: Use in ELISA.
Applications Tested: Tested as capture antibody in sandwich ELISA for human and mouse TGF-beta detection, combined with detection antibody eBio16TFB (Product #13-9923-81).
Recommended capture concentration: 6–10 µg/mL.
Include a standard curve (8000 pg/mL–60 pg/mL) in each ELISA plate.
This antibody recognizes the mature/active form of TGF-beta1 (not associated with LAP). Most samples require acid treatment and neutralization to remove LAP before assay. Test samples immediately after treatment; some serum/plasma may contain free TGF-beta1 measurable without acid treatment.
