Product Specific Information

This antibody detects endogenous levels of Smad4 only when phosphorylated at Thr277 (historically referenced as Thr276).

Target Information

Smad4 is encoded by the SMAD4 gene located on chromosome 18 in humans. It is also known as Smad family member 4, Sma- and Mad-related protein 4, or Mothers against decapentaplegic (MAD) homolog 4.
Smad4 contains N-terminal MH1 (MAD homology 1) and C-terminal MH2 (MAD homology 2) domains involved in DNA binding and protein interactions.
Upon activation by TGF-beta or BMP ligands, receptor-regulated Smads (R-Smads: 1, 2, 3, 5, 8) are phosphorylated, form heterotrimers with Smad4 (co-Smad), and translocate to the nucleus to regulate gene expression.
Smad4 acts as a tumor suppressor by modulating genes such as Cyclin D1 (downregulated) and collagen (upregulated), thereby inhibiting cell proliferation.
Dephosphorylation regulates its nuclear export and nucleocytoplasmic dynamics.
