Target Information
Janus kinase 3 (JAK3) phosphorylates members of the STAT (signal transduction and activators of transcription) family. JAK3 is predominantly expressed in immune cells and mediates signaling in response to interleukin receptor activation.
Upon cytokine stimulation (e.g., interferons, interleukins), JAKs become phosphorylated at adjacent tyrosine residues, activating downstream STAT proteins. Activated STATs dimerize, translocate to the nucleus, and regulate transcription of genes such as interferon-stimulated genes.
JAK2 is essential for IFN-γ receptor complex initiation, with JAK1 acting as an amplifier. Some studies suggest that Tyk2 and JAK3 may substitute for JAK2 under certain conditions within the receptor complex.
