Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
Suggested positive control: tonsil tissue.
Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family involved in signal transduction, T cell differentiation, and activation.
CD8 is mainly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with T cell receptor (TCR) for MHC class I molecules.
CD8α is essential for MHC-I binding and is also found on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8α homodimers.
MHC-I/peptide complex ligation recruits Lck kinase, leading to lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation.
Activated CTLs are critical for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
