Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
After retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is breast carcinoma or skin tissue.
Target Information
Cytokeratin 18 (Keratin 18, KRT18, type I cytoskeletal 18) is a low molecular weight acidic keratin that pairs with cytokeratin 8.
It is expressed in epithelial tissues of the gastrointestinal, respiratory, and urogenital tracts, as well as endocrine/exocrine and mesothelial cells.
Cytokeratin 18 is prevalent in non-squamous epithelia and adenocarcinomas, but absent in squamous cell carcinomas.
Hepatocellular carcinomas express only cytokeratins 8 and 18.
Mutations in the cytokeratin 18 gene are associated with cryptogenic cirrhosis.
Two transcript variants encoding the same protein are known.
Cytokeratin subtype expression patterns aid in distinguishing epithelial malignancies and are valuable in immunohistochemistry, cytopathology, and flow cytometry assays.
