Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes followed by cooling at room temperature for 20 minutes.
Following antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is tonsil or B cell lymphoma.
Target Information
CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa) expressed on mature and most malignant B cells, as well as a subset of T lymphocytes and follicular dendritic cells.
It regulates transmembrane calcium conductance, cell cycle progression, and B-cell proliferation.
CD20 is associated with lipid rafts, and its localization changes upon BCR aggregation.
CD20 remains on the cell surface after receptor ligation, making it a useful therapeutic target for B-cell depletion.
Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
