This antibody recognizes an extracellular epitope of CD40 (BP50), a 48 kDa type I single-chain transmembrane glycoprotein expressed on normal and neoplastic B cells, but not on terminally differentiated plasma cells. CD40 antigen is also present on Hodgkin’s and Reed-Sternberg cells, follicular dendritic cells, some macrophages, basal epithelial cells, and endothelial cells.
Target Information
The CD40 antigen is a single-chain glycoprotein belonging to the tumor necrosis factor/nerve growth factor superfamily. It shows significant homology to the Hodgkin’s disease-associated antigen CD30. CD40 is present on all B cells except plasma cells and is also found on some epithelial cells, carcinomas, and lymphoid dendritic cells.
CD40 plays a key role in immune and inflammatory responses, including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation. The transcription factor AKNA regulates CD40 expression, influencing homotypic cell interactions. TNFR2 adaptor protein interacts with CD40 to mediate signal transduction. The CD40–ligand interaction is implicated in amyloid-beta-induced microglial activation, an early event in Alzheimer’s disease pathogenesis. Two alternatively spliced transcript variants encoding distinct isoforms have been reported. Diseases associated with CD40 dysfunction include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
