Product Specific Information

Description:
The eBioC1.7 monoclonal antibody reacts with human CD244 (2B4, p38). CD244 is a 38 kDa protein expressed on NK cells, subsets of CD8+ T cells, γδ T cells, monocytes, basophils, and eosinophils. Binding of CD48, the CD244 ligand, activates NK cells, enhancing cytotoxicity and IFN-γ secretion. The C1.7 clone binds the same residue in the V domain as CD48, inducing activation through tyrosine phosphorylation and SAP recruitment. Mutations in SAP (as in X-linked lymphoproliferative disorder) prevent this interaction.

Reported for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometry of normal human peripheral blood cells. Recommended usage: 5 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Contains improved formulation over CellBlox. Required for optimal NovaFluor staining.

• Add 5 µL per sample (regardless of number of NovaFluors).
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor Conjugate Technology:

Target Information

CD244 (2B4, SLAMF4) is a cell surface receptor on NK cells and subsets of T cells mediating non-MHC restricted killing. Interaction with SLAMF2 modulates NK-cell cytolytic activity. Altered SLAMF4 expression is observed in autoimmune diseases such as systemic lupus erythematosus and rheumatoid arthritis. CD244 dysfunction is associated with lymphoproliferative syndromes. Multiple splice variants exist.
