Product Specific Information
The purified antibody detects a 65 kDa* protein corresponding to the apparent molecular mass of PDK1 on SDS-PAGE immunoblots of human MDA-MB-435 cells treated with EGF, but not in untreated cells.
PDK1 (Ser-241) reactivity was also observed by immunohistochemistry in human breast carcinoma tissue.
Target Information
PDPK1 (3-Phosphoinositide Dependent Protein Kinase 1) phosphorylates AGC kinases and activates conventional PKC and PKC zeta through phosphorylation of threonine residues in the activation loop.
It also phosphorylates Protein Kinase B (PKB) at threonine 308 in the presence of phosphatidylinositol-3,4,5-trisphosphate.
Active Akt inactivates Glycogen Synthase Kinase 3 (GSK3), leading to dephosphorylation and activation of glycogen synthase and stimulation of glycogen synthesis.
Because of its role in insulin-induced glycogen synthesis and PKC activation, PDPK1 is a potentially important target for metabolic drug research.
