Product Specific Information

The eBio1D3 (1D3) monoclonal antibody reacts with mouse CD19, a 95 kDa transmembrane glycoprotein expressed by B cells during all developmental stages except terminally differentiated plasma cells. Follicular dendritic cells also express CD19. Together with CD21, CD81, and MHC class II, CD19 forms a multimolecular complex associated with the B cell receptor (BCR), mediating signaling that induces tyrosine phosphorylation, calcium flux, and proliferation of B cells.

• 1 vial of CellBlox Plus Blocking Buffer

Applications

• Reported for use in flow cytometric analysis of mouse splenocytes.
• Tested at ≤ 0.1 µg per test (100 µL final volume, 10⁵–10⁸ cells/test).
• Optimal titration recommended for best performance.

Important Notes

• NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, or DAPI.
  Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance compared to the original CellBlox.
• Add 5 µL of CellBlox Plus Blocking Buffer per sample to antibody cocktails before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on novel nucleic acid dye structures engineered for optimized fluorescent signatures with minimized spillover and spread.

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. It is expressed on all peripheral B cells and certain myeloid leukemia cells. CD19 functions as a co-receptor with the BCR, lowering the activation threshold for antigen receptor signaling. It regulates normal and malignant B cell proliferation and is expressed in all B cell precursor leukemias.
Mutation of CD19 leads to hypogammaglobulinemia, while overexpression causes B cell hyperactivity.
