MA3-006 detects several members of the heat shock protein 70 kDa (HSP70) gene family, including HSP70, HSC70, p75, and HSP72 after heat shock, across various species such as yeast, Drosophila, fish, porcine, plant, mouse, avian, arthropod (red claw crayfish & black tiger prawn), amphibian, non-human primate, and human samples.
It has been successfully used in Western blot, immunocytochemistry, immunofluorescence, gel shift, and immunoprecipitation procedures.
By Western blot, this antibody detects proteins from approximately 70–78 kDa representing different members of the HSP70 family. Two-dimensional gel electrophoresis is required to resolve the heat-induced forms from constitutively expressed counterparts.
Immunocytochemical staining of HSP70 in heat-shocked HeLa cells results in cytoplasmic staining.
Epitope mapping suggests the recognized region is between amino acids 504–617 of human HSP70, involved in stress-induced nucleolar localization.
Target Information
HSP70 is a heat shock protein expressed in response to biological stresses such as high temperature.
Major HSP families include HSP70, HSP90, and HSP100. The HSP70 family consists of highly conserved proteins induced by various stresses in all examined organisms.
Human HSP70 family members include:
- HSP70: strongly inducible and constitutively expressed in primate cells
- HSP72: stress-induced 72 kDa protein
- HSC70: constitutively expressed 72 kDa cognate protein involved in clathrin-coated vesicle uncoating
- GRP78 (BiP): 78 kDa glucose-regulated protein localized in the endoplasmic reticulum
- p75 (HSP75): mitochondrial 75 kDa protein
HSP70 stabilizes existing proteins against aggregation, assists folding of newly translated proteins, and interacts with the ubiquitin-proteasome pathway.
The HSP70 gene is located in the MHC class III region, clustered with two related genes encoding similar proteins.
