Target Information
The c-Myc protein is a 62 kDa transcription factor encoded by the c-Myc gene located on human chromosome 8q24. It is commonly activated in various tumor cells and plays a key role in cellular proliferation, differentiation, apoptosis, and cell cycle progression.
Phosphorylation of c-Myc, particularly at Thr58/Ser62 by kinases such as glycogen synthase kinase 3, cyclin-dependent kinase, ERK2, and JNK, is associated with regulation of cell proliferation and cycle control. c-Myc is essential for vasculogenesis and angiogenesis during tumor development.
The c-myc oncogene (p62 c-myc) regulates normal cellular proliferation and differentiation, while deregulated expression can induce apoptosis. Antibodies against c-Myc epitopes recognize overexpressed proteins containing Myc epitope tags fused to either terminal of target proteins.
Amplification of the c-Myc gene has been observed in human tumors such as lung, breast, and colon carcinomas, whereas N-Myc amplification is found in neuroblastomas. Chromosomal translocations involving c-Myc (chromosome 8) and immunoglobulin loci (chromosomes 14, 2, or 22) are characteristic of Burkitt’s lymphoma and other B-cell lymphoproliferative disorders. Aberrant c-Myc expression is found in tumors of diverse origins including colorectal, gastric, hepatic, mammary, ovarian, pulmonary, and hematopoietic neoplasms.
