Product Specific Information

The 61D3 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, macrophages, and dendritic cells. CD14 binds LPS-LBP complexes with high affinity.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Pre-diluted and tested for flow cytometry at 5 µL (0.25 µg) per test
  • Recommended Use: 1 test = 100 µL final volume, suitable for 10⁵–10⁸ cells/test

Super Bright 780 is a tandem dye excited by the 405 nm violet laser and emits at 780 nm. Use with a 780/60 bandpass filter. Protect from light.

When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Compensation Notes:
Higher compensation values may occur in the violet 450/50 channel when using UltraComp eBeads microspheres (Product # 01-2222) compared to single-color stained cells. Compensation should be set using cells when possible. Similar results observed with AbC Total Antibody Compensation Beads (Product # A10497).

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal loss of brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It functions as a multifunctional lipopolysaccharide receptor and exists in both membrane-bound and soluble forms. CD14 binds LPS via LBP, contributing to immune recognition and inflammatory responses. Soluble CD14 can discriminate structural differences in LPS and modulate immune reactions. CD14 dysfunction is associated with infections and inflammatory diseases, including Mycobacterium chelonae infection and croup.
