Product Specific Information

The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), a low-affinity receptor for IgG (50–80 kDa).
CD16 includes two genes:

• CD16A (FcγRIIIA): expressed in macrophages and NK cells
• CD16B (FcγRIIIB): expressed in neutrophils

CD16 binds IgG1 and IgG3 preferentially and initiates signal transduction leading to ADCC, phagocytosis, degranulation, and proliferation.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry of normal human peripheral blood cells (5 µL/test).
A test is defined as the amount of antibody staining 10⁵–10⁸ cells in 100 µL final volume.

Fluorochrome Information:
Super Bright 780 is a tandem dye excited by 405 nm and emits at 780 nm. Recommended filter: 780/60 bandpass. Verify instrument compatibility.

Buffer Recommendation:
Use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Compensation Guidance:
Higher compensation values may occur in the violet 450/50 channel using UltraComp eBeads (Product # 01-2222-42) compared to stained cells. Set compensation with cells when possible.

Light Sensitivity:
Protect vial and samples from light due to photo-induced oxidation sensitivity.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product # 00-8222)
• 1-step Fix/Lyse Solution (Product # 00-5333)

CD16 encodes a receptor that recognizes the Fc portion of IgG and mediates immune complex clearance, ADCC, and viral infection enhancement.

• Gene: FCGR3A (chromosome 1)
• Expression: NK cells (membrane glycoprotein, transmembrane anchor)
• Related gene: FCGR3B (expressed in neutrophils, PI-anchored)
  Mutations in FCGR3A are associated with Immunodeficiency 20, recurrent viral infections, systemic lupus erythematosus, and alloimmune neonatal neutropenia.
