Product Specific Information

The YGIC86 monoclonal antibody reacts with mouse Glycoprotein A Repetitions Predominant (GARP, also known as Lrrc32 or Garpin). GARP is an approximately 80 kDa glycoprotein expressed on the cell surface. Expression has been reported in placenta, lung, kidney, heart, ovary, liver, skeletal muscle, and pancreas. Protein expression is observed on megakaryocytes, platelets, and activated regulatory T (Treg) cells. Expression of GARP on Treg cells is necessary for their suppressive function, possibly related to its role as a receptor for LAP/TGF beta.

For detecting GARP on platelets, use the Foxp3 Staining Buffer Set (Product #00-5523) to fix platelets before staining. Fixation/permeabilization is not necessary for Treg cells.

• Cross-reactivity: YGIC86 does not cross-react with rat GARP.
• Applications Reported: Flow cytometric analysis.
• Applications Tested: Tested on stimulated mouse splenocytes at ≤1 µg/test (in 100 µL final volume). Cell count: 10⁵–10⁸ cells/test. Optimize concentration as needed.
• Fluorophore: eFluor® 450 (excitation 405 nm, emission 445 nm; Violet laser).
• Filtration: 0.2 µm post-manufacturing filtered.

Target Information

GARP (leucine rich repeat containing 32) is a type I membrane protein with 22 leucine-rich repeats and a short intracytoplasmic region. It is expressed in CD4⁺, CD25⁺, FoxP3⁺ Treg cells and may serve as a marker for these cells.
