Product Specific Information
The HTA125 monoclonal antibody reacts with human Toll-like receptor 4 (TLR4). TLR4 is a type I transmembrane protein involved in innate immune responses. It forms a complex with MD-2 and CD14 to recognize and signal in response to bacterial lipopolysaccharides (LPS). TLR4 is primarily expressed on peripheral blood monocytes and at lower levels on granulocytes and immature dendritic cells.
- Recommended staining method: Lysed whole blood protocol for optimal TLR4 detection.
- Density gradient separation can reduce staining intensity.
- Applications Reported: Flow cytometric analysis.
- Tested Application: Flow cytometry using 5 µL (0.5 µg) per test (in 100 µL final volume).
- Suitable for detection of 10⁵–10⁸ cells/test.
- eFluor 450 emits at 446 nm and is excited by the 405 nm violet laser line.
- Ensure instrument compatibility with eFluor 450 fluorochrome.

Target Information
TLR4 (Toll-like receptor 4) is part of the TLR family responsible for pathogen recognition and activation of innate immunity. It is highly expressed in placenta and myelomonocytic leukocytes. TLR4 mediates responses to lipopolysaccharide (LPS) from Gram-negative bacteria by forming a receptor complex with CD14 and LY96 (MD-2). Mutations in TLR4 can alter LPS responsiveness. It is expressed in peripheral blood monocytes, a subset of B cells, and placenta.
