Product Specific Information

The L243 monoclonal antibody reacts with the human major histocompatibility complex (MHC) class II molecule, HLA-DR.
HLA-DR is expressed on antigen-presenting cells (APCs) including B cells, monocytes, macrophages, dendritic cells, and activated T cells. It is a heterodimeric transmembrane protein composed of alpha and beta subunits that play an essential role in presenting peptides to CD4⁺ T lymphocytes.

The L243 antibody recognizes a different epitope than LN3 and does not cross-block binding.

  • Tested Application: Flow cytometry using normal human peripheral blood cells
  • Recommended Use: 5 µL (0.125 µg) per 100 µL sample (10⁵–10⁸ cells/test)

eFluor® 450 is an alternative to Pacific Blue®, excited at 405 nm and emitting at 445 nm. Ensure the cytometer supports Violet laser detection.

Target Information

HLA-DR is a transmembrane glycoprotein composed of a 36 kDa alpha chain (DRA) and a 27 kDa beta chain (DRB).

  • The alpha chain gene (DRA) contains 5 exons:
    • Exon 1: leader peptide
    • Exons 2–3: extracellular domains
    • Exon 4: transmembrane domain and cytoplasmic tail
  • DRA acts as the sole alpha chain for DRB1, DRB3, DRB4, and DRB5.
  • The beta chain (DRB) carries polymorphisms that define peptide-binding specificity.
  • Hundreds of DRB1 alleles exist and are routinely typed in bone marrow and kidney transplantation.

HLA-DR is primarily expressed on antigen-presenting cells such as B lymphocytes, monocytes, macrophages, thymic epithelial cells, and activated T lymphocytes.
MHC class II molecules (DR, DQ, DP) bind intracellularly processed peptides, present them to T-helper cells, and play a critical role in immune response initiation.
