Protein disulfide isomerase (PDI) is a multifunctional enzyme that catalyzes the formation, breakage, and rearrangement of disulfide bonds.
- At the cell surface, PDI acts as a reductase, modifying exofacial proteins.
- Inside the cell, it assists in disulfide bond formation and rearrangement in nascent proteins.
- At high concentrations, it functions as a chaperone preventing aggregation of misfolded proteins, while at low concentrations it may promote aggregation.
- PDI also participates in hormone biogenesis and serves as a structural subunit in enzymes such as prolyl 4-hydroxylase and microsomal triacylglycerol transfer protein (MTTP).
- It interacts with LGALS9, retaining P4HB at the cell surface of Th2 T helper cells, enhancing disulfide reductase activity and cell migration.
