Product Specific Information

Description:
Monoclonal antibody HP-F1 recognizes CD85j (also known as ILT2, LILRB1, LIR1). CD85j is a single transmembrane glycoprotein with four ITIMs signaling through SHP-1. It is expressed on myeloid cells (monocytes, dendritic cells) and subsets of NK, T, and B cells. Expression correlates with leukemias (ALL, CLL) and viral infections (HIV, EBV, CMV). Ligands include MHC Class I molecules such as HLA-G, A, F, B27, E, and F. HP-F1 antibody reduces CD16-dependent cytolytic activity of NK cells.

Each product contains one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested for flow cytometry of normal human peripheral blood cells. Use 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before mixing with cells.
  • For single-color controls: 5 µL per 100 µL of cell sample.

Based on Phiton™ technology utilizing nucleic acid dye structures for engineered fluorescent signatures with reduced spillover and spread.

Target Information

This gene belongs to the leukocyte immunoglobulin-like receptor (LIR) family located at chromosomal region 19q13.4. The encoded protein has extracellular immunoglobulin domains, a transmembrane region, and cytoplasmic ITIMs. It binds MHC class I molecules on antigen-presenting cells and transduces inhibitory signals that regulate immune responses and cytotoxicity. Multiple transcript variants exist.
