Product Specific Information

The monoclonal antibody HP-F1 recognizes CD85j (ILT2), also known as LILRB1 and LIR1.
CD85j is a transmembrane glycoprotein with a long cytoplasmic domain containing four ITIMs that signal through SHP-1.
Expression is observed on myeloid cells (monocytes, dendritic cells) and certain lymphoid cells (NK, T, B subsets). It correlates with leukemias (ALL, CLL) and viral infections (HIV, EBV, CMV).

Ligands for CD85j include MHC Class I molecules (HLA-G, A, F, B27, E, F).
HP-F1 antibody reduces CD16-dependent cytolytic activity of NK cells.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells.
Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume).

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for optimal results.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) should be added to antibody cocktails before combining with cells:

  • Add 5 µL per sample (regardless of NovaFluor count)
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells)

NovaFluor conjugates utilize Phiton™ technology, engineered for minimal spillover and spectral spread.

Target Information

The CD85j (LILRB1) gene is part of the leukocyte immunoglobulin-like receptor (LIR) family located on chromosome 19q13.4.
It encodes a receptor expressed on immune cells that binds MHC class I molecules and transduces inhibitory signals to modulate immune responses, inflammation, and cytotoxicity.
Multiple transcript variants encode different isoforms.
