Product Specific Information

The monoclonal antibody B6H12 reacts with CD47, also known as integrin-associated protein (IAP) and neurophilin. CD47 is a glycosylated five-transmembrane protein involved in adhesion through interactions with SIRP and integrins (CD51/CD61, CD41/CD61). It mediates bi-directional signaling affecting neural synaptic activity and macrophage phagocytosis. CD47 acts as a receptor for thrombospondin and plays roles in T cell activation or apoptosis depending on thrombospondin levels.

This antibody has neutralizing activity and is supplied with NovaFluor conjugate and CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometry, immunoprecipitation, western blotting, immunohistochemistry (FFPE).
Applications Tested: Pre-titrated for flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (1.0 µg) per test (100 µL total volume). Recommended cell count: 10⁵–10⁸ cells/test.

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (PI, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining and should be added at 5 µL per sample or per 100 µL cell suspension.

NovaFluor conjugates utilize Phiton technology, enabling engineered fluorescence with minimal spillover and spread.
Excitation: 509 nm; Emission: 665 nm; Laser: 488 nm (Blue).

Target Information

CD47 (Integrin-associated protein, IAP) is a transmembrane glycoprotein interacting with integrins and modulating signaling pathways, including G protein activation and calcium influx during cell adhesion. It serves as a receptor for thrombospondin and regulates membrane transport and signal transduction. CD47 is widely expressed and interacts with CD172a (SIRPα) to inhibit macrophage phagocytosis. It also influences cell migration, B cell adhesion, T cell activation, chondrocyte response to mechanical signals, and neuronal development.
