Target Information

CDC34 accepts ubiquitin from the E1 complex and catalyzes its covalent attachment to other proteins. It catalyzes Lys-48-linked polyubiquitination and cooperates with E2 UBCH5C and the SCF (FBXW11) E3 ligase complex for polyubiquitination of NFKBIA, leading to its proteasomal degradation.
CDC34 participates in ubiquitin chain elongation, working with UBE2D3 as the initiator E2. It also cooperates with the SCF (SKP2) E3 ligase complex to regulate cell proliferation through ubiquitination and degradation of MYBL2 and KIP1.
Additionally, it is involved in degradation of CREM isoform ICERIIgamma, ATF15, WEE1 kinase, and beta-catenin. CDC34 is also a target of human herpes virus 1 protein ICP0, leading to ICP0-dependent interaction with proteasomes.
